Overview
Lexicon is developing small molecules intended to treat ocular diseases such as glaucoma. As part of the Genome5000 project, Lexicon scientists identified a kinase known as LG710 that is responsible for regulating intraocular pressure (IOP). IOP levels can become elevated due to increased resistance in the conventional fluid outflow system in the eye, a mechanism that accounts for 90% of the total drainage in the human eye. Compounds targeting LG710 are intended to lower IOP by increasing conventional fluid outflow. The goal of the LG710 program is to develop a once-daily eye drop that lowers IOP by increasing conventional fluid outflow in the eye, a novel mechanism not currently addressed by approved products.

Preclinical Data
In preclinical studies, topical application of LG710 compounds decreased IOP in a disease model of ocular hypertension. Backup compounds have also been identified; medicinal chemistry efforts and formulation optimization are in progress.
